Since the introduction of curse seals to the Naruto series, people have been wondering how it works and why it works the way it does. So let's cut the crap and get right to it.

Firstly we all know that the curse seal was created by Orochimaru's tampering with a certain enzyme. This enzyme was special and naturally produced in the body of a prisoner he held, a man named Jugo. Jugo had bouts of rage in which he would lose total control of himself, often strange scales and enlargements of limbs occur as a result.

The Enzyme

What is an enzyme first of all. An enzyme is a molecule that helps catalyze reactions at a sub molecular level. Essentially it helps speed up chemical reactions. For example Chemical A and Chemical B come into contact, and they can react. If you add an enzyme, it can greatly speed up the reaction. In some cases, some chemicals need an enzyme to properly react with one another.

The History behind Life

Human's have only been around for approx. 1, 000, 000 years. Now this time frame does not include the first appearance of the modern human, us, Homo Sapiens But long, long, and I say long before even the appearances of humans, and even the prehistoric mammals, or even the dinosaurs, there were bizarre creatures that lurked the earth. The earth was still covered mostly in water. The first time signs of life started appearing, was exclusively water-borne. The earliest complex creatures were arthropods (simply insects) that resided in the massive oceans. They all had exo-skeletons. Actually the earliest sign of our true ancestors were a bottom jaw-less fish species. It was the first creature to ever develop an internal spine, in short, a skeletal system. So how does this tie in with humans? The fish were inferior creatures to the arthropods at the time. So they always had to escape, because they were always prey. They way they reproduced was by jumping into an isolated inland pool of water where they must go on land for a very short amount of time. So in time, specifically several hundreds of millions of years, they developed limbs adaptable to land movement. And in time, a primitive lung, a labyrinth lung to be exact, to take air. Several million years pass, and land creatures came to be. Evolution does not stop. The need for creatures to be able to control and maintain body temperature spawned the warm-blooded mammals. Early mammalian ancestors actual looked a lot more like reptiles and were gigantic, compared to the small furrier creatures we know today. So now we have the cold blooded, the warm blooded, and the insects. All of these creatures were at one point in time inter related. And as evolution has gone by, the DNA used to program to build these animals were forever lost. Lost, meaning that somewhere in the human genetic code, some where, where it is extremely hidden, we still have the simple codes for these creatures. Of course keep in mind that a lot of this is still scientific theory, and needs to be proven.

The Conclusion

So how do all these scientific principles tie in with Naruto's curse seal? This is my theory. Jugo's body had a terribly and extremely rare genetic mutation, in which his DNA had programmed his human body to produce this unknown enzyme. This enzyme in return sporadically catalyzes the hidden "prehistoric" DNA to fuse with his current and normal Human DNA. Depending on which "prehistoric" DNA was triggered, it would produce various results. Orochimaru extracted this enzyme, and he himself has done many other alterations to the enzyme make up. He altered the way it behaved, but in the end, the essential function is to extract a piece of that "prehistoric" DNA to fuse with the normal human DNA.

This can be clearly seen in Kimimaro and Sasuke. Two diffrent seals, namely Earth and Heaven. You can see that Kimimaro reverts to a very dinosaur like form and Sasuke gains webbed wings, similar to that of a reptiles. There are many other strange transformations. I won't name any specific people, but examples include: extra limbs, toughened skin, exo-skeletons etc. I had also mentioned that only simple parts of the prehistoric creature's DNA are kept, this is the reason why their whole body does not change entirely into a creature.

The darkened and changed skin colour of the user is triggered by the mixed DNA in which it has told the body to produce specific appendages that coincide with the "prehistoric" DNA that specific enzyme (curse seal) has extracted. The darkened skin is caused by irregular and increased metabolic levels, which explain a very quick appendage growth and enhanced healing. The effects are much more terrible on Jugo, since he is the original Enzyme producer. It's too effective on his body, to the point where it drives him to go on random rage episodes, because his brain processes have been replaced by that of prehistoric, primal ones; essentially thinking based on pure primal instincts.

So what does this mean? More cellular parts in the body= more chakra. More chakra= heightened fighting abilities. This technique is obviously not without its draw backs. Increased metabolism in the body will result in higher bodily temperatures. Staying in state like this for too long will ruin the homeostasis processes (state of balance) of the body. This is why it is so tiring and painful to constantly call on the curse seal for help.

Orochimaru and the Cure

Without a doubt, Orochimaru has a cure. Although this cure probably doesn't eradicate the enzyme production, but merely supresses it. Orochimaru has injected himself with the enzyme obviously, so that he could transmit it to his test subjects. Although why he doesn't just inject them, himself, I'm not quite sure. The enzyme alteration might actually occur inside himself. His body might actually be the one that varies each bite and injection of the enzyme, ultimately producing different curse seals. Let's also not forget that he can probably control it, seeing as how he's given two of the same curse seals to two separate people (Anko and Sasuke) But he's gone now, kind of. He resides in Kabutos head. I think he still knows where the cures are located or how to synthesize them.

That's my theory, hope you liked it. I tried to make it as short as possible without losing valuable info. I just tried to explain this as much as I possibly could to make the most sense of how these affected people go all weird and animal-ish. ALSO The history and enzyme portion of my theory, is very, very, very condensed. If you are interested in learning more I suggest you go to an encyclopedia or wikipedia. I also highly recommend that you read BBC's Walking With series, it's amazing and informative of prehistoric life origins.

I think genetically, we're 50% the same as a Banana. Around 75% the same as earthworms. I think it's easier to say that he has some weird gene that makes his body mutate weird and get more chakra.

Adding real-life genetics into Naruto just convolutes it.

but there is not only curse seal can do such transformations - Naruto's natural energy transforms hin into a frog(wich is not ancient creature)
