no it was only a filler moment in the anime not manga, and buggy was just so suprised that rayleigh helped him and that luffy has such an infamous family that he thought rayleigh was also his uncle which is not true (keep in mind buggy is a clown)

Uncle is an honorific term people in Japan use for elders that are close to them; sort of like how Capone's men call him father. It doesn't have to mean they are literally uncle and nephew.
