If he was trying to commit suicide, yes. Obviously he has to consciously use healing techs, they dont just automatically activate every time he gets hurt. If he was actually trying to die, one kunai stab could do the job.

Raikage is using lightning to strengthen his body's defense. Without it it is possible he could be stabbed too. After all steel is harder than human skin.
In case of Hahirama, he doesn't have as tough of a body as third raikage, but he does have super human healing ability because of his Senju dna. If he could voluntarily stop his body from healing his wounds, for example a kunai thru the heart, then he can die from blood lost and cardiac failure.

He can heal without seals, but it's still a technique that requires him to activate it. Also, we don't know if he had the healing technique at that time or if he developed it later. Even for Hashirama it's easy to die from a simple wound if you don't prepare and don't block or dodge it.
