Simply look in someone's eyes and right there the genjutsu is activated at an instant or does itachi have to look into someone eyes and then activate it which delays it ? Which is it ? How fast does tsukoyomi even activate ?

Tsukyomi moves at the speed of light, because it requires your eyesight to work. So yes, it's almost practically instant.

tsukuyomi isnt 'instant', all it does is manipulate the victim's perception of time inside the mind [1], hence it is 'instant' cause there is no chance for an outside partner to help you.
