      I think yes, cuz during fight between Sasuke and Itachi , Itachi used tsukomi to grab sasukes eyes, he didn't knew it was an illusion - he acted like he acted in reality and if Sasuke couldn't resist , he would take sasukes eyes

    itachi was planning to take sasuke's eyes but sasuke was stronger than him
    Itachi wanted to kill tobi after he obtained sasuke's eyes

    No, I think Tobi is quite serious about what hes talking about. If he was lying then he would admit it to Sasuke, face-to-face. But I really believe what Tobi is trying to say to make Sasuke understand why the genocide of the Uchiha Clan happened.
